My motherboard is Intel Corporation D865GLC AAC27499-405, with a Pentium 4 and about 2Gb RAM. Can I replace my current hard drives with capacities as large as 250 Gb?

It all depends. You need Windows XP with Service Pack 2 or 3 to install more than 132 GB in one partition. Otherwise, you will have to create multiple partitions if your install disk has SP 1, or no service pack.
If your install disk has SP2 or SP3, you can use hard drives of 750 GB or even 1 TB.
